A well-run workshop does not just build things; it also clears out broken, worn, or no-longer-needed equipment, otherwise it would drown in junk. Cells are the same. They constantly need to destroy old, damaged, or unwanted proteins, both to recycle their parts and to keep the place clean. The proteasome is the cell's main protein-shredding machine for doing exactly that.

The proteasome is a large, barrel-shaped protein complex with a hollow core. Proteins marked for destruction are fed into one end, unfolded and threaded inside, and chopped into short fragments by enzymes lining the interior chamber; those fragments are then recycled back into individual amino acids. Crucially, the proteasome is selective — it does not shred just anything that floats by. Proteins are first tagged with a chain of a small marker protein called ubiquitin, and the proteasome's 'cap' recognizes that tag, grabs the doomed protein, and feeds it in. The whole hollow design keeps the destructive enzymes safely walled off so they can only act on proteins deliberately delivered inside.

The proteasome matters because controlled protein destruction is just as important as protein construction. It removes misfolded or damaged proteins before they can clump and cause harm, and it ends the life of regulatory proteins on schedule — which is how the cell, for instance, advances cleanly from one stage of division to the next by destroying specific proteins at the right moment. Because it sits at so many control points, the proteasome is a major drug target; some cancer treatments work by inhibiting it, overwhelming cancer cells with their own undegraded proteins.

To leave one phase of the cell cycle and enter the next, a cell destroys specific timing proteins called cyclins by tagging them with ubiquitin and feeding them to the proteasome. Stop that destruction and the cell's division clock jams — proof that timely shredding is as vital as building.

Destroying the right protein at the right moment keeps the cell cycle ticking forward.

The proteasome handles individual tagged proteins; bulk cleanup of whole organelles or large clumps is the job of lysosomes and autophagy instead. The two systems are complementary, not interchangeable.
