permissive effect

A permissive effect is when one hormone does not directly cause a response itself, but is required for another hormone to work fully. Think of it as someone holding the door open: they are not the one walking through, but without them the second person could not get in. The first hormone sets the stage so the second can act.

The classic example involves thyroid hormone and the catecholamines. Thyroid hormone does not by itself drive the heart-racing, fat-mobilizing effects of epinephrine, but it increases the number of beta-adrenergic receptors, so epinephrine produces a much stronger effect when thyroid hormone is present. Cortisol is also broadly permissive — it maintains the responsiveness of blood vessels to catecholamines, which is partly why a severe cortisol deficiency can cause dangerously low blood pressure.

The key distinction is that the permissive hormone is necessary but not sufficient: it enables the action without producing it. This is different from synergism, where both hormones actively contribute to the same effect. Recognizing permissiveness explains why some hormone deficiencies cause trouble not by their own absent action but by leaving another system unable to respond.
