Hormone antagonism is when one hormone opposes the action of another, pulling a regulated value in the opposite direction. It is like having both an accelerator and a brake: by playing two opposing controls against each other, the body can hold a value steady and adjust it quickly in either direction. The balance between the two, not either one alone, sets the result.

The textbook pair is insulin and glucagon in glucose control. Insulin lowers blood glucose by promoting its uptake and storage, while glucagon raises it by releasing glucose from the liver; the ratio between them, shifting with feeding and fasting, governs blood sugar. Calcium balance has a similar opposing arrangement, with PTH raising blood calcium and calcitonin (in animals more than humans) tending to lower it.

Antagonism is valuable precisely because opposing forces give fine, fast, two-way control. It also explains certain disease patterns: in type 1 diabetes the loss of insulin leaves glucagon's glucose-raising action unopposed, contributing to the runaway high blood sugar and ketone production of diabetic ketoacidosis.
