Picture a doorway into a workshop, and right beside it a small bracket where a guard can plant himself. When the guard stands there, he physically blocks the doorway and no one gets in to start work; when he steps away, the door is clear. The operator is that little bracket built into the DNA — a short, specific stretch of sequence that a regulatory protein can sit on to control whether work begins.
Concretely, the operator is a short DNA sequence located at or right next to the promoter of an operon. A regulatory protein — most famously a repressor — recognizes the operator's exact sequence and binds to it tightly, the way a hand fits a specific molded grip. Because the operator overlaps or sits just downstream of where RNA polymerase needs to settle, a protein bound there blocks the polymerase from starting transcription, like a parked car blocking a driveway. Remove or release that protein and the polymerase is free to begin. The operator itself does not code for any protein; it is purely a control site, a recognizable address where a regulator docks.
The operator is the physical heart of negative control in bacteria — the actual piece of DNA that gets occupied or vacated to switch an operon off or on. It is where the abstract idea of a 'gene switch' becomes a concrete molecular handshake between a protein and a precise sequence. Studying operators taught biologists how proteins read DNA sequence-specifically, a recognition logic that runs through all of gene regulation.
In the lac operon, the lac repressor protein clamps onto the operator that overlaps the promoter. With the repressor parked there, RNA polymerase cannot move forward, so the operon stays off until something pries the repressor loose.
The operator is a docking address for a regulator, not a gene that codes for protein.
Despite the name, the operator is not a gene and codes for nothing — it is a DNA binding site. Do not confuse it with the regulatory gene, which is a separate gene that encodes the protein that binds the operator.
