How does RNA polymerase know where a gene begins, out of millions of base pairs that all look chemically similar? It reads a road sign. The promoter is that sign — a specific stretch of DNA, sitting just before a gene, that says 'start transcribing here, on this strand, in this direction.'

A promoter is a DNA sequence near the start of a gene that RNA polymerase (or its helper proteins) recognizes and binds. It does two jobs at once: it marks the start point, and because it has a defined orientation it also dictates which strand is template and which way transcription runs. Promoters are not transcribed into the useful part of the RNA — they are docking and instruction sequences. Their exact shape differs between life's domains. Bacterial promoters are compact, built around two short conserved motifs (the -10 and -35 boxes, named for their distance upstream of the start) recognized directly by the polymerase's sigma factor. Eukaryotic promoters are more elaborate, often featuring a TATA box and serving as a landing platform where many general transcription factors assemble before the polymerase joins.

The promoter is where the decision to express a gene is largely made and tuned. A 'strong' promoter is bound readily and fires often, giving lots of RNA; a 'weak' one fires rarely. Regulatory proteins (activators and repressors) and other DNA elements (enhancers, operators) work by changing how well the polymerase engages the promoter. Because promoters set both the location and the rate of transcription, they are central to gene regulation — and a favourite tool in genetic engineering, where swapping in a strong promoter can crank up the output of an introduced gene.

Biotech labs put a target gene behind a strong viral promoter so a bacterium or cell makes the encoded protein in large amounts — the same promoter logic the cell uses, borrowed for manufacturing.

Promoter strength sets how much RNA — and thus protein — a gene yields.

A promoter is not the same as an enhancer. The promoter sits right at the start and is where transcription begins; an enhancer can sit far away (even thousands of bases off) and merely boosts the rate. Both shape expression, but only the promoter marks the start site.
