Some crystals invite water molecules to move in and become permanent residents of the structure, sitting at regular spots within the lattice. A crystal that incorporates water this way is a hydrate. The water is not just surface dampness — it is built into the crystal's repeating pattern, like raisins baked evenly through a loaf rather than sprinkled on top.
A hydrate is a specific kind of solvate, the general term for a crystal that traps solvent molecules. The amount of water is usually a fixed ratio to the drug, giving named forms such as a monohydrate or dihydrate. Because the water occupies lattice positions, a hydrate is a genuinely different solid form from the anhydrous (water-free) crystal, with its own melting point, stability, and, importantly, solubility — anhydrous forms often dissolve faster than their hydrates.
Hydrates matter because a drug can gain or lose its lattice water as humidity changes, switching between forms during manufacturing or storage and altering its dissolution behavior. A formulation developed around the anhydrous form can underperform if the drug quietly converts to a slower-dissolving hydrate on the shelf. Controlling and monitoring the hydration state is therefore a routine but critical part of solid-form development.
Lattice water differs from absorbed surface moisture: lattice water sits at defined positions and leaves only at characteristic temperatures, whereas surface moisture comes and goes freely with humidity.
