Think of two strands of rope twisted tightly around each other. Before you can use either rope on its own, you have to untwist them and pull them apart — and doing that takes effort. Helicase is the molecular machine that does exactly this to DNA: it grips the double helix, unwinds the two coiled strands, and separates them, opening the way for copying to happen.

Helicase is an enzyme that clamps around the DNA, usually encircling one strand, and motors along it. It breaks the weak hydrogen bonds that hold the two strands together — the bonds between paired bases — so the double helix splits into two single strands. This work is not free: helicase burns chemical fuel (it hydrolyzes ATP) to power its movement, much as an engine burns fuel to do mechanical work. Sitting right at the very front of the replication fork, helicase is the lead vehicle, opening the DNA so all the copying machinery behind it has single strands to read.

Helicase matters because DNA's two strands cannot be copied while they remain zipped together; the bases that carry the information are tucked on the inside of the helix and must be exposed. Helicases also work outside of replication — in repair, in transcription, and in untangling RNA — so the family is large and ancient. A common misconception is that the strands simply 'fall apart' on their own; at body temperature the double helix is quite stable, and it takes a dedicated, fuel-burning machine to pry it open at the right place and the right speed.

At a fast-moving bacterial replication fork, helicase unwinds the helix at around a thousand base pairs per second — like a zipper being yanked open so fast the eye can barely follow. The single strands it exposes are instantly grabbed by other proteins so they cannot zip back up.

Helicase is the fuel-burning machine at the very front of the fork.

Helicase only separates the strands; it does not copy them. And unwinding ahead of it builds up twisting strain that another enzyme, topoisomerase, must relieve.
