GLUT2
GLUT2 is the glucose-sensing window of the beta cell. Unlike the insulin-controlled GLUT4, GLUT2 is always open, and it has a low affinity for glucose, meaning it moves glucose proportionally to how much is in the blood. This turns the beta cell into an honest glucose meter: the more sugar outside, the more flows in.
Because glucose entry through GLUT2 keeps pace with blood levels, the rate of glucose metabolism — and therefore ATP production — inside the beta cell tracks blood glucose closely. This is the first step that links a rising blood sugar to insulin release, feeding into the ATP-driven chain that ends in granule secretion. GLUT2 also serves a sensing role in the liver and parts of the kidney and gut.
A caveat worth noting: in humans the beta cell may rely more on other transporters (such as GLUT1 and GLUT3) than rodent beta cells do, so the textbook GLUT2-centric picture comes largely from animal studies. Nonetheless, the principle — a non-saturating transporter that lets the cell read ambient glucose — is what makes glucose sensing possible.
