人人都知道,细胞里的信息是 DNA → RNA → 蛋白质这样流动的。后来,两个实验室发现了一种把它倒着运行的酶。

核心想法

生物学的「中心法则」说:遗传信息只朝一个方向走——从 DNA,被抄写成 RNA,再被读取来建造蛋白质。反过来似乎是不可能的——没有任何已知的东西,能把 RNA 重新变回 DNA。

可是 1970 年,两位各自独立工作的科学家,发现的恰恰就是这样一种东西:一种藏在某些病毒体内的酶,能读取一条 RNA 链,写出一条与之匹配的 DNA 链。他们称它为逆转录酶。信息原来真的可以 RNA → DNA 地流动——而一整个家族的病毒,竟都依赖着它。

它是如何诞生的

多年来,霍华德·特明一直坚持:一种只由 RNA 构成的病毒,必定会以某种方式给自己造出一份 DNA 拷贝,并把这份拷贝悄悄塞进宿主细胞自己的基因组里——一个隐藏的「前病毒」。多数生物学家都认为这想法是错的,因为它需要一个谁也没见过的反向步骤。

特明(与水谷哲)以及完全另起炉灶的戴维·巴尔的摩,都去寻找那缺失的酶——结果两边都在病毒颗粒内部找到了它。他们的两篇论文,并排出现在 1970 年 6 月同一期《自然》上。结果干净利落,几周之内便被重复并接受。1975 年,特明与巴尔的摩同雷纳托·杜尔贝科分享了诺贝尔奖。

它为何重要

它改写了一条人人都学过的规则,也开创了一个领域。使用逆转录酶的那些病毒,如今被称为逆转录病毒——而其中一种,在十年后被发现,就是 HIV。正是因为理解了逆转录,科学家才得以设计出最早对抗艾滋病的药物:那些卡住这种酶、让病毒无法复制自己的药。

一个可以想象的画面

想象一台只朝一个方向运转的印刷机:用一块金属母版(DNA)印出纸质拷贝(RNA),再用纸做出产品(蛋白质)。逆转录酶,就是一台能拿一张纸质拷贝、反过来浇铸出一块全新金属母版的机器——让印刷机倒着走。一种逆转录病毒到来时,只带着「纸」,然后自己造出母版,再把它悄悄归档进你的母版库里。

它的位置

这是对克里克 1958 年中心法则的重大修正,也是沃森与克里克双螺旋的续篇。它开创了逆转录病毒学——十年后,正是这门学问让人们读懂了 HIV——也交给分子生物学一件最常用的工具,即 RT-PCR 里的那个 RT。而维持我们自己染色体末端完整的那种酶——端粒酶,结果也是一种逆转录酶。

In the issue of 27 June 1970, Nature carried two independent reports — one from Baltimore at MIT, one from Temin and Mizutani at Wisconsin — that reached the same conclusion, and grouped them under the heading "Viral RNA-dependent DNA Polymerase."
The question
Since the mid-1960s Temin had argued, against the consensus, that RNA tumour viruses such as Rous sarcoma virus replicate through a DNA intermediate — a "provirus" integrated into the host chromosome. That demanded an unknown step: copying RNA into DNA, a direction no enzyme was known to take and which seemed to run against the central dogma.
The experiment
Purified virions were gently disrupted with a detergent and incubated with the four deoxyribonucleoside triphosphates and a magnesium salt. The virions synthesized DNA on their own. Crucially, treating the virions with ribonuclease beforehand abolished the synthesis — showing that the template being copied was the viral RNA — and the product behaved as DNA and hybridized back to that RNA.
The finding
Each virion carries its own enzyme, an RNA-dependent DNA polymerase — later named reverse transcriptase — that transcribes the viral RNA genome into DNA. Genetic information can flow RNA → DNA. The result was reproduced within weeks and accepted almost at once.
