核心想法

基因是用一套四个字母的字母表写成的(RNA 中的碱基 A、U、G、C),但生命体是由蛋白质构成的,而蛋白质是 20 种不同氨基酸连成的链。所以,一定存在一套密码——一种把 RNA 的字母翻译成氨基酸的办法。人人都相信这套密码真实存在,可到 1961 年,还没有人读出过它的哪怕一个词。

尼伦伯格与马特伊,找到了读出一个词的办法。他们自己写出一条最简单的讯息:一条只由一个字母 U 一遍遍重复构成的 RNA。当他们把这条「poly-U」喂给一台细胞的造蛋白机器时,它造出的蛋白质,竟也是由唯一一种氨基酸——苯丙氨酸——一遍遍重复构成的。讯息「UUU」的意思,就是「苯丙氨酸」。密码的第一个词,被读出来了。

它是如何诞生的

到 1961 年,编码问题已是新生的分子生物学中那个最大的未解之谜,许多绝顶聪明的人,写满了一本本笔记本,构想出种种巧妙的密码——而其中绝大多数是错的。马歇尔·尼伦伯格,一位年轻、默默无闻、供职于美国国立卫生研究院的科学家,选了另一条路:做实验。他与德国生物化学家海因里希·马特伊一道,搭起一套「无细胞」系统——把细菌打破后取出的、仍能运转的内部机器,养在试管里,依旧能造出蛋白质。

1961 年 5 月 27 日凌晨,马特伊做了那支决定性的试管:细胞汁液、各种原料,再加一条纯由 U 构成的合成链。它迸发出一阵苯丙氨酸蛋白质。当那年夏天尼伦伯格在莫斯科的一次大会上宣布此事时——起初台下几乎空无一人——这消息令整个领域为之一震。一套曾让最聪明的理论家们束手无策的密码,竟被一个直截了当的实验破解了。

它为何重要

它把遗传密码,从一个想法,变成了可以一个字母、一个字母去读的东西。方法一旦确立,密码余下的部分便迅速破译,给了我们那部通用字典——把 DNA 翻译成构建一切生命的蛋白质。这正是现代遗传学、医学与生物技术的根基。

一个可以想象的画面

想象你捡到一条很长的加密讯息,却没有密钥。于是你「作弊」:自己写一条只含字母「U」、重复上千遍的讯息,塞进解码机里。出来的译文,是只有「苯丙氨酸」这个词、重复上千遍。现在你确知了一件事——「U」(每三个一组)的意思,是「苯丙氨酸」。再用一整页纯「A」、纯「C」如法炮制,一次一项,你便能把整把密钥重建出来。生命的密码,最初正是这样被读出来的。

它的位置

沃森与克里克(1953)揭示了 DNA 的结构;克里克的中心法则(1958)说,信息沿 DNA → RNA → 蛋白质流动。但二者都没说字母「如何」翻译。这个实验,补上了那部缺失字典的第一项;短短几年内,整套密码便为人所知——这正是基因与蛋白质之间的纽带,也是今天读取、编辑基因的一切所赖以建立的。

The system
The paper works with a cell-free extract from Escherichia coli — broken-open cells that still supply ribosomes, transfer RNA, enzymes and an energy source — in which protein synthesis can be driven in a test tube. Adding an RNA template to this extract makes it incorporate radioactively labelled amino acids into newly made protein, which is then measured.
The synthetic templates
Instead of natural messenger RNA of unknown sequence, the authors added synthetic polyribonucleotides of known, simple composition — including polyuridylic acid (poly-U), a chain built from a single repeated base, U.
The decisive result
Poly-U specifically and dramatically stimulated the incorporation of one amino acid — phenylalanine — producing polyphenylalanine, while other amino acids were not. Because poly-U is nothing but a long run of U's, this tied a run of uridine to phenylalanine and, read as triplets, pointed to UUU as a codon for phenylalanine: the first entry of the genetic code to be deciphered.
What followed
The same approach soon assigned further codons — poly-A directing lysine, poly-C directing proline — and the complete 64-codon table was finished within a few years.
