In depth · the introduction

The question: nervous or chemical?
The paper opens on a known fact — that acid entering the small intestine provokes a flow of pancreatic juice — and on the prevailing explanation, descended from Pavlov's school, that this was a nervous reflex carried along the nerves of the gut.
The decisive experiment
Bayliss and Starling isolated a loop of jejunum in an anaesthetised dog and cut every nerve to it, leaving it connected only by its blood vessels. They introduced dilute acid into the loop — and the pancreas, at a distance, still poured out juice. A nervous reflex was ruled out: the signal had to travel by blood.
Secretin
They then scraped the lining of the jejunum, ground it with sand and dilute acid, filtered the result, and injected the filtrate into a vein. The pancreas responded as vigorously as to acid in the gut itself. The active substance — released from the gut wall into the blood by acid — they named secretin.

Conclusion
The authors conclude that pancreatic secretion is governed not by a nerve but by a chemical messenger produced in the intestinal lining and carried in the bloodstream. The full paper — with its seventeen figures and secretion tracings — is available at the source below.
