人人都知道,細胞裡的資訊是 DNA → RNA → 蛋白質這樣流動的。後來,兩個實驗室發現了一種把它倒著運行的酶。

核心想法

生物學的「中心法則」說:遺傳資訊只朝一個方向走——從 DNA,被抄寫成 RNA,再被讀取來建造蛋白質。反過來似乎是不可能的——沒有任何已知的東西,能把 RNA 重新變回 DNA。

可是 1970 年,兩位各自獨立工作的科學家,發現的恰恰就是這樣一種東西:一種藏在某些病毒體內的酶,能讀取一條 RNA 鏈,寫出一條與之匹配的 DNA 鏈。他們稱它為逆轉錄酶。資訊原來真的可以 RNA → DNA 地流動——而一整個家族的病毒,竟都依賴著它。

它是如何誕生的

多年來,霍華德·特明一直堅持:一種只由 RNA 構成的病毒,必定會以某種方式給自己造出一份 DNA 拷貝,並把這份拷貝悄悄塞進宿主細胞自己的基因組裡——一個隱藏的「前病毒」。多數生物學家都認為這想法是錯的,因為它需要一個誰也沒見過的反向步驟。

特明(與水谷哲)以及完全另起爐灶的大衛·巴爾的摩,都去尋找那缺失的酶——結果兩邊都在病毒顆粒內部找到了它。他們的兩篇論文,並排出現在 1970 年 6 月同一期《自然》上。結果乾淨俐落,幾週之內便被重複並接受。1975 年,特明與巴爾的摩同雷納托·杜爾貝科分享了諾貝爾獎。

它為何重要

它改寫了一條人人都學過的規則,也開創了一個領域。使用逆轉錄酶的那些病毒,如今被稱為逆轉錄病毒——而其中一種,在十年後被發現,就是 HIV。正是因為理解了逆轉錄,科學家才得以設計出最早對抗愛滋病的藥物:那些卡住這種酶、讓病毒無法複製自己的藥。

一個可以想像的畫面

想像一台只朝一個方向運轉的印刷機:用一塊金屬母版(DNA)印出紙質拷貝(RNA),再用紙做出產品(蛋白質)。逆轉錄酶,就是一台能拿一張紙質拷貝、反過來澆鑄出一塊全新金屬母版的機器——讓印刷機倒著走。一種逆轉錄病毒到來時,只帶著「紙」,然後自己造出母版,再把它悄悄歸檔進你的母版庫裡。

它的位置

這是對克里克 1958 年中心法則的重大修正,也是沃森與克里克雙螺旋的續篇。它開創了逆轉錄病毒學——十年後,正是這門學問讓人們讀懂了 HIV——也交給分子生物學一件最常用的工具,即 RT-PCR 裡的那個 RT。而維持我們自己染色體末端完整的那種酶——端粒酶,結果也是一種逆轉錄酶。

In the issue of 27 June 1970, Nature carried two independent reports — one from Baltimore at MIT, one from Temin and Mizutani at Wisconsin — that reached the same conclusion, and grouped them under the heading "Viral RNA-dependent DNA Polymerase."
The question
Since the mid-1960s Temin had argued, against the consensus, that RNA tumour viruses such as Rous sarcoma virus replicate through a DNA intermediate — a "provirus" integrated into the host chromosome. That demanded an unknown step: copying RNA into DNA, a direction no enzyme was known to take and which seemed to run against the central dogma.
The experiment
Purified virions were gently disrupted with a detergent and incubated with the four deoxyribonucleoside triphosphates and a magnesium salt. The virions synthesized DNA on their own. Crucially, treating the virions with ribonuclease beforehand abolished the synthesis — showing that the template being copied was the viral RNA — and the product behaved as DNA and hybridized back to that RNA.
The finding
Each virion carries its own enzyme, an RNA-dependent DNA polymerase — later named reverse transcriptase — that transcribes the viral RNA genome into DNA. Genetic information can flow RNA → DNA. The result was reproduced within weeks and accepted almost at once.
