A gradient is a ruler
A morphogen is a signaling molecule that spreads from a source and forms a concentration gradient — high near the source, low far away. Cells measure how much morphogen they sense and use that level as a coordinate, like reading a ruler. High concentration might mean ‘head end,’ medium ‘middle,’ low ‘tail end.’ Different thresholds switch on different transcription factors, so one smooth gradient is carved into sharp bands of gene expression.
The mother gets there first
In the fly, the very first gradients are set up before the embryo makes any of its own gene products. The mother deposits messenger RNA from maternal-effect genes at specific ends of the egg. One such gene, *bicoid*, is anchored at the future head end; its protein diffuses to form a head-to-tail gradient that acts as a morphogen, telling cells which end is which.
Maternal-effect genes have a strange inheritance signature: the offspring's phenotype depends on the mother's genotype, not the embryo's own, because the mother stocked the egg in advance. A mother lacking working *bicoid* lays eggs that fail to make a head — even if the embryo itself carries a good copy. This was a vital clue that the body axis is set up by maternally supplied gradients.
From gradient to pattern, step by step
- The mother loads maternal-effect gene mRNAs at the egg's poles, defining front and back before fertilization is even complete.
- Their proteins diffuse into smooth morphogen gradients along the egg.
- Concentration thresholds switch on the embryo's own ‘gap’ and ‘pair-rule’ genes in coarse, then finer, stripes — progressively subdividing the body.
- Those stripes hand off to the homeotic / Hox genes from the previous guide, which give each segment its final identity.
This is gene regulation working as a relay: each layer of genes reads the pattern made by the layer before and sharpens it. The smooth maternal gradient becomes coarse bands, then fine stripes, then segment identities — a continuous signal refined into a precise, repeatable body plan.
