Reading and writing both have a direction — you read English left to right, not right to left. Nucleic acid chains have a direction too, and transcription obeys it strictly. Understanding that direction explains why RNA is built one way while the polymerase scans the template the opposite way, like a typist reading a page from the bottom up while typing from the top down.

Every nucleic acid strand has two chemically different ends, called 5' and 3' (named after carbon positions on the sugar). RNA polymerase can only add a new nucleotide to the free 3' end of the growing RNA — so the RNA is always synthesized in the 5'-to-3' direction. Because the template strand is antiparallel (runs the opposite way), the polymerase must read the template in the 3'-to-5' direction. Picture two lanes of traffic going opposite ways: the polymerase walks along the template from its 3' end toward its 5' end, and as it goes it lays down RNA growing from 5' toward 3', the newest nucleotide always at the leading edge. One more vital fact: RNA polymerase can initiate a fresh chain on its own — it needs no primer, unlike DNA polymerase, which can only extend an existing 3' end.

This fixed direction is not a mere bookkeeping detail. It defines where a gene 'starts' (its 5' end, the promoter side) and where it 'ends', it sets which strand can serve as template for a given gene, and it underlies the convention of writing sequences 5'-to-3'. The same chemistry that makes the 3' end the only place new nucleotides can attach governs replication and translation too, giving the whole flow of molecular information a consistent grain.

Template read 3'-to-5': 3'-T A C-5'. RNA grows 5'-to-3': the polymerase adds A (opposite T), then U (opposite A), then G (opposite C), giving 5'-A U G-3' — the start codon spelled out as it is made.

RNA is synthesized 5'->3' while the template is read 3'->5'; the two run antiparallel.

A common slip is to say the polymerase 'moves 5'-to-3'.' Be precise: the RNA grows 5'-to-3', but the enzyme moves along the template toward the template's 5' end (i.e., reads 3'-to-5').
