template versus coding strand

DNA is double-stranded, like a zipper with two halves. When the cell transcribes a gene, it does not copy both halves — it only reads one of the two strands. That raises a natural question: which strand, and why does the other one matter at all? The answer is that the two strands play very different roles, and biologists give them different names: the template strand and the coding strand.

The template strand is the one RNA polymerase actually reads. Because RNA is built by base-pairing, the new RNA comes out complementary to this template — wherever the template says A, the RNA gets U, and so on. The coding strand is the other strand, the one that is not read. Here is the elegant part: since the RNA is complementary to the template, it ends up matching the coding strand letter for letter (just with U swapping in for T). That is exactly why it is called the coding strand — its sequence reads the same as the messenger RNA, and so it 'spells out' the eventual protein code that scientists usually write down.

Two cautions keep people from getting tangled. First, which strand is the template is not fixed for the whole chromosome — it depends on the gene. One gene may use the top strand as its template while a neighboring gene uses the bottom strand, depending on which way each gene points. Second, the choice is not arbitrary: the promoter's orientation determines which strand is read, ensuring the RNA comes out with the correct, meaningful sequence rather than gibberish.

If the template strand reads 3'-TAC-5', RNA polymerase builds the RNA 5'-AUG-3'. Notice that this AUG is identical to the coding strand (which reads 5'-ATG-3'), with U simply standing in for T — which is why textbooks quote the coding strand to show a gene's 'message'.

The RNA copies the template but reads out the same as the coding strand (with U for T).

The coding strand is the one whose sequence matches the mRNA, yet it is not the strand actually read — the template (its complement) is. Mixing these up is one of the most common transcription mistakes.
