structural alert

A structural alert is a particular chemical substructure that has been linked, across many compounds, to a heightened risk of toxicity. Think of it as a watch-list pattern: when chemists spot the substructure in a new molecule, they raise a yellow flag and look more closely.

These alerts are usually fragments that can undergo metabolic activation into reactive species, or that react directly with biological molecules. Familiar examples include anilines, nitroaromatics, certain Michael acceptors, hydrazines, and thiophenes, which can be oxidized into electrophiles that bond to proteins or DNA. The danger is rarely the intact group itself but what the body's enzymes turn it into.

A structural alert is a flag, not a verdict. Many safe, marketed drugs contain alerting groups whose risk is offset by low dose, fast clearance, or blocked metabolism. The right response is to investigate, for example by trapping reactive metabolites or testing for genotoxicity, and to redesign the molecule only if a real liability is confirmed.
