Picture a sprawling set of flattened sacs stacked like pita bread, spreading out from the nucleus, with thousands of tiny dots glued to their outer surface. Those dots are ribosomes, and the dotted, bumpy look gives this organelle its name. The rough endoplasmic reticulum is the cell's protein-finishing and packaging workshop, especially for proteins headed out of the cell or into membranes.

The rough ER is a network of membrane-bound flattened sacs (cisternae) that is continuous with the nuclear envelope and studded with bound ribosomes. As a ribosome on its surface builds a protein, the growing chain is threaded directly into the interior of the ER. Inside, the protein folds into shape, gets quality-checked, and often receives sugar tags (glycosylation). Properly made proteins are then pinched off in small membrane bubbles (transport vesicles) and sent toward the Golgi apparatus for further processing.

The rough ER is therefore the first station of the secretory pathway — the assembly line that produces proteins for export, for the cell surface, and for organelles like lysosomes. Cells specialized in secretion, such as the antibody-making cells of your immune system or the digestive-enzyme cells of your pancreas, are packed with rough ER, which is a direct visual sign of their job.

An insulin-producing cell in the pancreas is crammed with rough ER, because every insulin molecule it exports must first be folded and checked there before leaving the cell.

Secretory cells are visually dominated by rough ER — their export factory.

The ribosomes on rough ER are not permanently fixed there; they dock only while making a protein destined for the ER, and detach when finished.
