RNA interference

Imagine a busy mailroom where, to stop a particular message from being acted on, a guard simply intercepts and shreds every envelope that matches a known address. RNA interference is the cell's version of this targeted shredding. It is a system that uses small pieces of RNA to find and silence matching messenger RNAs — the working copies of genes — so the protein those messages encode never gets made, or gets made far less.

Precisely, RNA interference is a gene-silencing process triggered by short double-stranded RNA. The cell chops such RNA into tiny fragments (around 21-23 letters long), and one strand is loaded into a protein complex called RISC. This guide strand lets RISC scan the cell's messenger RNAs and recognize any whose sequence matches; the complex then cuts that messenger RNA or blocks its translation, shutting the gene's output down. Because the matching is by base-pairing, RNAi is exquisitely specific — it acts only on messages that read like its guide.

RNA interference matters in two big ways. In nature it is a defense and regulatory system: it silences viruses and roaming genetic parasites and helps fine-tune normal gene activity (closely related to microRNA control). In the lab and clinic it is a revolutionary tool: scientists can switch off almost any chosen gene by feeding in matching small RNA, which lets them probe gene function, and a small but growing number of approved drugs use this principle to silence disease-causing genes. The discovery earned a 2006 Nobel Prize.

An approved RNAi drug silences a gene in the liver that drives a rare nerve disease: a small synthetic RNA homes in on that gene's messenger RNA and has it destroyed, so the harmful protein is no longer produced.

RNAi turns genes down by attacking their RNA after it's made — it does not edit or delete the DNA, so its silencing is reversible and dose-dependent, unlike a permanent gene knockout.
