process validation
Before an airline lets a new plane carry passengers, it does not just fly it once; it proves the plane will perform reliably under defined conditions. Process validation does the same for a manufacturing process: it is the documented collection of evidence proving that a process, operated within set parameters, consistently produces a product meeting its predetermined specifications and quality attributes.
Modern regulatory thinking (the FDA 2011 guidance and ICH Q8–Q12) frames validation as a lifecycle in three stages. Stage 1, process design, defines the process from development and risk knowledge. Stage 2, process qualification, confirms the designed process can reproducibly perform at commercial scale — historically by making several consecutive successful batches (often three) under full monitoring. Stage 3, continued process verification, keeps watching routine production data to confirm the process stays in control over its life.
The key idea is consistency, not a single good batch: anyone can get lucky once, so validation demands repeatable evidence and built-in process understanding. Two honest points: validation is not a one-off certificate but an ongoing commitment, and it must be redone or extended after significant changes — a new site, scale, supplier or equipment can invalidate the earlier proof. Cleaning and analytical methods are validated by the same logic, separately.
To qualify a new tablet process, a company manufactures three consecutive full-scale batches under intensive sampling, tests blend uniformity, hardness, dissolution and assay on each, and shows all results sit comfortably within specification before routine production begins.
Several consecutive in-spec batches are the classic evidence of process consistency.
Process qualification has a hardware counterpart in equipment qualification — installation, operational and performance qualification (IQ/OQ/PQ) — which confirms the machines themselves are installed and run as intended before the process is validated on them.
