Imagine the difference between fresh-cut flowers and a houseplant that just keeps growing. Cells in culture come in two main flavours that work much like this. Primary cells are taken fresh from a living tissue and behave like the real thing, but, like cut flowers, they survive only a limited time and stop dividing. Cell lines are cells that have been changed so they divide indefinitely — a houseplant that never dies — but in becoming immortal they have drifted from a fully normal cell.

Primary cells are freshly isolated from a tissue and grown directly. They closely resemble the cells in the body, which makes them realistic, but normal cells can only divide a limited number of times — a ceiling called the Hayflick limit — after which they stop and eventually die. Cell lines are populations that have escaped that limit, either because they came from a tumour or because they were deliberately altered to keep dividing forever. They are convenient, endlessly available, and uniform — but the very changes that made them immortal often mean their genes and behaviour no longer perfectly match a healthy cell.

Choosing between them is a constant trade-off between realism and convenience. Primary cells give results closer to true biology but are scarce, variable, and short-lived; cell lines are easy, consistent, and cheap but can mislead because they are subtly abnormal. The famous HeLa line, derived from cancer cells, has grown in labs worldwide since 1951. A common pitfall is treating any old cell line as a stand-in for normal cells of that tissue, when it may have accumulated many genetic changes.

For a quick, repeatable screen a lab uses an immortal cell line that grows forever; but to confirm a finding reflects real biology, they repeat the key experiment on hard-to-get primary cells taken straight from tissue.

An immortal cell line is not a perfect copy of a normal cell: the genetic changes that let it divide forever (often cancer-related) can make its behaviour differ from healthy tissue, so findings should be confirmed in primary cells.
