A supersaturated solution is holding more dissolved drug than it really should — like a crowd squeezed past the room's official capacity. It looks calm and clear, but it is living on borrowed time: nudge it, and the excess crashes out as crystals.
Supersaturation is a metastable state in which the dissolved concentration exceeds the equilibrium solubility. It usually arises by changing conditions faster than crystals can form — cooling a hot solution, evaporating solvent, shifting pH, or diluting a cosolvent or amorphous formulation in the gut. The extra dissolved drug raises the concentration gradient that drives absorption, which is exactly why formulators try to create it.
The catch is instability. Because the system is above its true solubility, it tends to relieve the strain by nucleating and growing crystals, dropping back to equilibrium. So-called supersaturating drug delivery systems use precipitation inhibitors — often polymers such as HPMC or PVP — to slow nucleation and keep the drug supersaturated long enough to be absorbed: the deliberate strategy nicknamed “spring and parachute”, where the spring generates supersaturation and the parachute sustains it.
