p53
p53 is often called the guardian of the genome, and a good picture is a vigilant safety inspector standing at the factory door. Whenever a cell takes on too much DNA damage or stress, p53 steps in: it can halt the production line for repairs, or, if the damage is too severe, order the faulty cell to shut itself down for good.
Technically, p53 is the protein made by the TP53 gene, a tumor-suppressor that acts as a transcription factor. When it senses problems, it switches on other genes that pause the cell cycle, drive DNA repair, or trigger programmed cell death. This stops a damaged cell from copying its errors into daughter cells.
TP53 is the most commonly mutated gene across human cancers; when it is disabled, cells with dangerous DNA changes survive and divide. Inheriting one faulty TP53 copy causes Li-Fraumeni syndrome, a hereditary condition with high lifetime risk of several cancers. The inspector is powerful, but it is not a cure-all, and losing it is a major step toward malignancy.
The protein is p53; the human gene is written TP53. Its name comes from an early measured size of about 53 kilodaltons.
