orphan receptor
An orphan receptor is a receptor in search of its messenger. Scientists have found the protein — often by spotting a gene that resembles known receptors — but no one yet knows which natural molecule in the body normally switches it on. The receptor is real and structured like its relatives, but its 'parent' signal is missing, hence the name.
Orphans were discovered in large numbers once gene sequencing let researchers recognize receptor-shaped proteins from their DNA alone, far faster than the slow chemistry of finding each one's natural activator. The reverse hunt that follows — taking a known receptor and searching for the molecule that binds it — is called reverse pharmacology or 'deorphanization'.
When the matching ligand is finally identified, the receptor is 'adopted' and often becomes a fresh drug target, sometimes revealing an entirely new signalling system. Several blockbuster drug families grew out of formerly orphan receptors. The honest caveat is that some receptors stay orphans for decades, and a few may have no single dedicated ligand at all.
The cannabinoid receptors began as orphan GPCRs; identifying their endogenous ligands (the endocannabinoids) opened an entire field of drug research.
