Optical crosstalk & off-target excitation
Unintended activation that arises when light meant for one purpose or place also drives an opsin elsewhere. Two forms dominate. Spectral crosstalk: the imaging light (or a second opsin's light) overlaps the actuator's broad absorption band and spuriously spikes cells — the central obstacle to all-optical interrogation. Spatial / optical off-target excitation: scattered or out-of-focus light, together with opsin present in passing axons and dendrites, activates neighbors of the intended cell.
Managing crosstalk is the defining engineering of optical interfacing: red-shifted opsins spectrally separated from green indicators, soma-targeting to suppress neuropil drive, two-photon confinement, the minimal necessary light dose, and careful indicator/actuator pairing. Crosstalk is never fully eliminated, so honest all-optical work measures and reports residual off-target activation rather than assuming perfect specificity.
