Before a single amino acid can be added, the cell faces a setup problem: it must place the ribosome on the mRNA at exactly the right spot, in exactly the right reading frame, with the first tRNA already in position. Get this wrong and the whole protein is wrong. Initiation is this careful assembly of the machine on the starting line.

Initiation brings three things together at the start codon: the small ribosomal subunit, the mRNA, and a special initiator tRNA already charged with methionine. Helper proteins called initiation factors orchestrate the assembly, and the special initiator tRNA is loaded straight into the P site (not the A site), where the start codon AUG waits. Once everything is correctly lined up, the large subunit joins to complete the ribosome, and the machine is poised with its first amino acid in place and the A site empty, ready for the second tRNA. How the ribosome finds the start codon differs by domain of life: bacteria use a short Shine-Dalgarno sequence just upstream of AUG that base-pairs the small subunit into place, while eukaryotes attach near the mRNA's 5' cap and scan along the message until they reach the first good AUG.

Initiation is the slowest and most regulated step of translation, and for good reason: it is the cell's last chance to decide whether to make this protein at all and to be sure the reading frame is correct. Because it is rate-limiting, it is a prime control point — cells dial protein production up or down largely by tuning initiation, and many viruses hijack a cell by seizing its initiation machinery. The bacterial and eukaryotic versions differ enough that some antibiotics block initiation in bacteria without touching us.

In E. coli: the 30S subunit, guided by its Shine-Dalgarno match, settles on the mRNA so that the start AUG sits in the P site; initiator fMet-tRNA pairs there; then the 50S subunit clamps on to form the complete 70S ribosome, ready to elongate.

Small subunit + mRNA + initiator tRNA assemble at the start codon, then the large subunit joins.

Uniquely, the initiator tRNA loads into the P site, whereas every later tRNA enters through the A site. Initiation is also where translation is most heavily controlled — it is the cell's main volume knob for protein synthesis.
