When you start unzipping a long zipper, you have to begin somewhere. A cell copying a whole chromosome cannot just start anywhere at random — it begins at specific marked spots in the DNA called origins of replication. Think of them as the official 'start copying here' signs written into the sequence.
An origin is a particular stretch of DNA, often rich in A-T base pairs (which are held by only two hydrogen bonds and so are easier to pull apart than the three-bond G-C pairs). Initiator proteins recognize and bind the origin, pry the strands apart to make a small opened region, and recruit the rest of the replication machinery. From that opened spot, copying spreads outward in both directions, creating a replication bubble. A bacterium such as E. coli has a single origin on its one circular chromosome (called oriC). Eukaryotes, with vastly more DNA on multiple linear chromosomes, have thousands of origins firing along each chromosome so the whole genome can be finished in a reasonable time.
Origins matter because they set where and when copying starts, and that timing is tightly controlled so the genome is duplicated once and only once per cell cycle. Bacterial origins are well-defined sequences; in many eukaryotes, especially mammals, origins are less defined by a strict sequence and more by chromatin context, which is still an active research area. Mis-firing or re-firing of origins can cause parts of the genome to be over- or under-copied — a hallmark of genome instability and cancer.
E. coli's whole 4.6-million-base-pair circle is copied from one origin, oriC, with two forks racing in opposite directions until they meet on the far side. A human chromosome instead lights up many origins so its much longer DNA finishes within the few hours of S phase.
One bacterial origin versus thousands of eukaryotic origins.
Having one origin works for a small circular chromosome but would be far too slow for a giant eukaryotic genome — multiple origins are a necessity of scale, not a luxury.
