Michaelis-Menten kinetics (Km and Vmax)

Imagine a single ticket booth at a busy event. When only a few people show up, the booth serves them as fast as they arrive — more people, faster service. But once a long line forms, the booth is already working flat out; adding more people to the line cannot speed it up, because the bottleneck is the one clerk. Enzymes behave the same way, and Michaelis-Menten kinetics is the simple math that describes this.

As you raise the substrate concentration, the reaction rate rises steeply at first, then levels off toward a ceiling called Vmax — the maximum speed, reached when essentially every enzyme active site is occupied and working as fast as it can (saturation). Km, the Michaelis constant, is the substrate concentration at which the rate is exactly half of Vmax. Km is a rough measure of how tightly the enzyme grips its substrate: a low Km means the enzyme reaches half-speed even at low substrate levels (a firm, eager grip), while a high Km means it needs a lot of substrate to get going (a looser grip). Vmax tells you the enzyme's top speed; Km tells you how easily it gets there.

These two numbers let biologists compare enzymes, understand why a pathway speeds up or slows down, and predict how drugs that block enzymes will behave. The model is a deliberate simplification — it assumes one substrate, a steady state, and no product buildup — so real cells, with multiple substrates and regulation, are more complex. Still, Km and Vmax are among the most useful single facts you can know about an enzyme. A common confusion: a low Km means high affinity, which feels backwards until you remember it is the substrate level needed to reach half-speed — less needed means a tighter grip.

Two versions of an alcohol-processing enzyme can have very different Km values. A low-Km version starts clearing alcohol from the blood even at low concentrations, while a high-Km version only kicks in strongly when alcohol levels are high — part of why people process alcohol at different rates.

Vmax is the top speed; Km is the substrate level needed to reach half that speed.
