Hormone synthesis is the biochemical manufacturing process by which an endocrine cell makes its signaling molecule. Just as different factories need different assembly lines, different hormone classes are built in completely different ways, and the synthesis route shapes how the hormone is stored, released, and regulated.

Peptide and protein hormones are synthesized like any protein: genes are read into messenger RNA, ribosomes translate them into chains, and the chains are trimmed and folded before storage. Steroid hormones take a different route, built step by step from cholesterol by a series of enzymes (steroidogenesis) located in mitochondria and the endoplasmic reticulum. Amine hormones are made by chemically modifying a single amino acid.

Synthesis is a key control point in endocrinology. For peptide hormones, the cell can hold a large pre-made reserve and adjust gene transcription over the longer term. For steroids and thyroid hormones, output depends on the speed of enzyme reactions, so regulation often works by changing how active those enzymes are. Disorders of single synthesis enzymes — such as the blocks in congenital adrenal hyperplasia — show how a missing step can reshape an entire hormone profile.
