A candidate profile is the scorecard, written before optimization, that defines exactly what a molecule must achieve to be allowed into development. It is like a job description posted before interviewing: rather than judging applicants by gut feeling, you agree in advance on the must-have qualifications so everyone aims at the same bar.

The profile lists concrete targets for each important property: a minimum potency and selectivity, acceptable solubility and permeability, a target metabolic stability and predicted human half-life, clean safety screens such as no major hERG or genotoxicity flag, and a feasible synthetic route. Properties are often tiered into strict 'must-haves' and softer 'nice-to-haves', so the team knows which numbers are non-negotiable.

Setting the profile up front keeps optimization honest and focused, preventing teams from falling in love with a potent but otherwise flawed molecule. It is not frozen forever; as biology and the competitive landscape become clearer, the profile is sometimes revised. But a project without a clear candidate profile tends to wander, optimizing whatever is easiest rather than what actually matters.
