building block
A building block is a small, commercially available molecule that you bolt onto others to assemble a target, much like a LEGO brick. Rather than synthesizing every fragment from scratch, medicinal chemists order these pieces from catalogs containing tens of thousands of options and combine them with a handful of reliable reactions. The molecule you want emerges from snapping the right bricks together.
Useful building blocks carry a reactive handle suited to a dependable coupling, such as a boronic acid for a Suzuki reaction, an amine or carboxylic acid for an amide bond, or an aryl halide for a metal-catalyzed coupling. Because so many analogs differ only in which building block occupies one position, ordering a varied set lets a team explore structure–activity relationships quickly and cheaply.
The trade-off is that relying on what is purchasable can quietly bias a project toward easily made, look-alike molecules and away from novel chemical space. Some building blocks are also expensive, unstable, or available only in one enantiomer, so synthetic accessibility becomes a real design constraint, not just a convenience.
